Currently in Phase 2 clinical trials for moderate-to-severe atopic dermatitis, with potential expansion into eosinophilic esophagitis (EoE) and epidermal differentiation disorders (EDD). (Therapeutic antibody – biologic) - **TRIV-573** – A second-generation bispecific antibody combining dual inhibition of KLK5/7 with an anti-IL‑13 mechanism. Currently in Phase 1, with plans to initiate a Phase 2 proof-of-concept trial in moderate-to-severe atopic dermatitis in the second half of 2026. (Therapeutic antibody – bispecific) - **TRIV-920** – A first-in-class monoclonal antibody targeting trypsin 1 and 2 for the treatment of pancreatitis (preclinical stage). (Therapeutic antibody – first-in-class) ## Market Standing - **Valuation**: Not disclosed - **Key Metric**: Total funding of $223.4M (3 rounds: Series A $16.4M in 2020, Series A‑2 $92M in 2023, Series B $115M in 2024, and an undisclosed Series C led by Janus Henderson Investors and Ascenta Capital in 2026) - **Notable Investors/Partners**: Atlas Venture, Cormorant Asset Management, Goldman Sachs Alternatives, Janus Henderson Investors, Ascenta Capital - **Growth Signals**: 43.6% year-over-year headcount growth (now 47 employees); expanded into new headquarters in Watertown, MA; advanced TRIV‑509 into Phase 2; initiated first human studies for TRIV‑573; raised Series C to broaden clinical studies and drive company growth. ## Competitive Advantages - **Genetics‑informed target selection**: Uses integrated multi‑omic datasets and “knowledge graphs” to identify disease drivers (e.g., KLK5/7 in atopic dermatitis) and leverage both risk and protective alleles. - **Advanced antibody engineering**: Developed a single monoclonal antibody that binds both KLK5 and KLK7 with equal potency, overcomes historic targeting challenges, and includes half‑life extension for convenient dosing. - **First‑in‑class potential**: TRIV‑920 addresses pancreatitis, a condition with no approved therapies, showing preclinical reduction of pancreatic damage markers. ## Strategic Focus - Advance TRIV‑573 through Phase 2 proof‑of‑concept in atopic dermatitis (2H 2026). - Expand TRIV‑509 into additional indications (EoE, EDD). - Continue preclinical development of TRIV‑920 for pancreatitis. - Scale the organization while maintaining a collaborative, patient‑focused culture. ## Why Work Here - **Culture**: Described as “bold & rigorous,” collaborative, and kind, with a strong emphasis on patient impact. Employees note that leadership actively listens and creates a safe space for ideas. - **Location**: On-site in Watertown, MA (newly expanded headquarters) with an additional office in Waltham. - **Work Environment**: Clinical‑stage biotech with exciting science; opportunity to “make medicines for patients in need alongside colleagues pushing the boundaries of science and medicine.” - **Perks**: Not explicitly detailed, but the organization values “curious minds” and “drive to turn ideas into impact.” ## Sources 1.
